Identifying and Distinguishing Single-molecule Based on the Fluorescence Photon Statistic Measurement

Peng Shuang-Yan, Huang Tao

Identifying and distinguishing single-molecule by fluorescence photon statistic measuring have been widely applied in physics, biology, chemistry and information processing et al.. Two kinds of methods of identifying and distinguishing single-molecule are analyzed and reviewed. Firstly, two identification methods to identify singlemolecule photon source are introduced: one by measuring laser stimulated single-molecule fluorescence field twoorder autocorrelation function with start-stop method, the other based on single photon counting statistic measurement. Secondly, the other kind of methods are to distinguish which species molecules the single-molecule detected belongs to when a couple of species molecules are mixed. Two methods, based on property-correlated single-photon counting statistic measurement and probability calculation and analysis, are chiefly introduced: the maximum likelihood method and the lookup table method.

Quantum dynamics of individual molecules studied by photon distribution in blinking fluorescence

Игорь Сергеевич Осадько

Various methods of photon counting in single molecule blinking fluorescence are considered and photon distribution functions, , , and related to these methods are derived. It is shown that each exponential process exp(−λ j t) in quantum dynamics of a single molecule manifests itself in the mathematical expressions for the photon distribution function via Poissonian function PN (−λ j t) with the same rate constant. This relation enables one to avoid N-fold integration in the mathematical expressions for the functions, , , and derived in previous works. Numerical calculations of the functions , , and for blinking fluorescence are carried out. A relation between photon distribution and photo-electric pulse distribution is discussed. Thanks to relation found we can use photo-electric pulse distribution functions for studying photon distribution function and for studying quantum dynamics of single nanoparticle with blinking fluorescence.
