Abstract

We suggest an approach to detect the conformation of single molecule by using the photon counting statistics. The generalized Smoluchoswki equation is employed to describe the dynamical process of conformational change of single molecule. The resonant trajectories of the emission photon numbers $$ and the Mandel's $Q$ parameter, in the space of conformational coordinates $\bm{\mathcal{X}}$ and frequency $ω_L$ of external field ($\bm{\mathcal{X}}-ω_L$ space), can be used to rebuild the conformation of the single molecule. As an example, we consider Thioflavin T molecule. It demonstrates that the results of conformations extracted by employing the photon counting statistics is excellent agreement with the results of {\it ab initio} computation.

Photon statistics in single molecule orientational imaging

Matthew R. Foreman, Sherif S. Sherif, Péter Török

Optical techniques in single molecule imaging rely heavily on photon counting for data acquisition. Extraction of information from the recorded readings is often done by means of statistical signal processing, however this requires a full knowledge of the photoelectron statistics. In addition to counting statistics we include a specific form of random signal variations namely reorientational dynamics, or wobble to derive the general probability density function of the number of detected photons. The relative importance of the two factors is dependent upon the total number of photons in the system and results are given in all regimes.

Theory of single molecule emission spectroscopy

Golan Bel, Frank L. H. Brown

A general theory and calculation framework for the prediction of frequency-resolved single molecule photon counting statistics is presented. Expressions for the generating function of photon counts are derived, both for the case of naive "detection" based solely on photon emission from the molecule and also for experimentally realizable detection of emitted photons, and are used to explicitly calculate low-order photon-counting moments. The two cases of naive detection versus physical detection are compared to one another and it is demonstrated that the physical detection scheme resolves certain inconsistencies predicted via the naive detection approach. Applications to two different models for molecular dynamics are considered: a simple two-level system and a two-level absorber subject to spectral diffusion.

Photon Counting Statistics for Single Molecule Spectroscopy

Eli Barkai, YounJoon Jung, R. Silbey

We present a general relation between Mandel's $Q$ parameter describing variance of number of photons emitted from a single molecule and a three time correlation function describing a spectral diffusion process the molecule is undergoing. For a prototype of such a stochastic process an exact solution is found which exhibits rich types of physical behaviors.

Single-Molecule Photon Counting Statistics via Generalized Optical Bloch Equations

Yujun Zheng, Frank L. H. Brown

We derive the generating function for single molecule photon emission events within the context of the stochastically modulated optical Bloch equations. Statistical properties of single molecule photon counting experiments are deduced from a set of coupled differential equations only slightly more complicated than the Bloch equations themselves. This formulation allows for the study of photon bunching and antibunching within a single theoretical framework and provides a description of these behaviors that emphasizes the connection with single molecule experiments. Application is made to the spectroscopy of a chromophore coupled to a single two level system.

Single Molecule Photon Counting Statistics for Quantum Mechanical Chromophore Dynamics

Golan Bel, Yujun Zheng, Frank L. H. Brown

We extend the generating function technique for calculation of single molecule photon emission statistics (Zheng, Y.; Brown, F. L. H. Phys. Rev. Lett. 2003, 90, 238305) to systems governed by multi-level quantum dynamics. This opens up the possibility to study phenomena that are outside the realm of purely stochastic and mixed quantum-stochastic models. In particular, the present methodology allows for calculation of photon statistics that are spectrally resolved and subject to quantum coherence. Several model calculations illustrate the generality of the technique and highlight quantitative and qualitative differences between quantum mechanical models and related stochastic approximations when they arise. Calculations suggest that studying photon statistics as a function of photon frequency has the potential to reveal more about system dynamics than the usual broadband detection schemes.

Statistics of emission photon from single molecule

Yujun Zheng

The emission photon statistics from single molecule based on the generating function method developed recently is discussed. From the view of statistical theory, the generating function could be thought as the partition function of the “counting field s”. The statistical properties of emission photon from single molecule are also discussed.
